Product Intro:
Dyronib is a targeted therapy used primarily in hematological malignancies, specifically Philadelphia chromosome-positive (Ph+) leukemias. It blocks the abnormal proteins that drive cancer cell growth and division, offering an effective option when other treatments fail or resistance develops.

Uses (Indications):
  1. Chronic Myeloid Leukemia (CML) – chronic phase
  2. CML – accelerated or blast crisis phase
  3. Acute Lymphoblastic Leukemia (ALL) – Philadelphia chromosome-positive (Ph+)
  4. For use in adult and pediatric patients as prescribed
  5. Can be used in patients who are resistant or intolerant to other TKIs
  6. Sometimes used off-label in clinical trials for solid tumors expressing certain mutations

Storage Instructions:
  1. Store at room temperature between 20°C to 25°C
  2. Protect from moisture and light
  3. Keep out of reach of children
  4. Do not use tablets if they are broken or show signs of damage

How It Works (Mechanism of Action):
Dasatinib inhibits BCR-ABL tyrosine kinase and other kinases responsible for uncontrolled cell division in leukemia. By blocking these kinases, it stops the proliferation of cancerous white blood cells and promotes normal hematopoiesis.

Side Effects:
Common Side Effects:
  1. Headache
  2. Diarrhea
  3. Fatigue
  4. Nausea and vomiting
  5. Skin rash
  6. Low platelet and red blood cell counts
Serious Side Effects:
  1. Pleural effusion (fluid in the lungs)
  2. QT prolongation (abnormal heart rhythm)
  3. Myelosuppression (decrease in bone marrow activity)
  4. Liver enzyme elevation
  5. Pulmonary arterial hypertension (rare but serious)
  6. Bleeding complications due to low platelet count

Dosage (Typical Recommended Dose):
  1. CML (chronic phase): 100 mg once daily
  2. CML (accelerated/blast phase) or Ph+ ALL: 140 mg once daily
  3. Pediatric dose: Based on body surface area (mg/m²), under physician supervision
  4. Dose adjustments may be needed based on tolerance, side effects, or lab values
  5. Always take as prescribed by the oncologist

Method of Administration:
  1. Take orally, with or without food
  2. Swallow the tablet whole with water (do not crush or chew)
  3. Take at the same time every day for best results
  4. If vomiting occurs after taking, do not retake the dose — follow up with your doctor

Precautions:
  1. Baseline and periodic blood counts are essential
  2. Monitor for signs of fluid retention or shortness of breath
  3. Avoid use in pregnancy – may harm the fetus
  4. Use effective contraception during and for 30 days after treatment
  5. Avoid grapefruit or grapefruit juice (can increase drug levels)
  6. Monitor liver function and heart rhythm during treatment

Drug Interactions:
  1. CYP3A4 inhibitors (e.g., ketoconazole, erythromycin) – increase dasatinib levels
  2. CYP3A4 inducers (e.g., rifampin, phenytoin) – reduce effectiveness
  3. Antacids and proton pump inhibitors – reduce absorption of dasatinib
  4. Antiplatelet and anticoagulant drugs – increased bleeding risk
  5. Drugs that prolong QT interval – increase risk of heart arrhythmias

Allergies (Warnings for Allergic Reactions):
  1. Hypersensitivity reactions are rare but may occur
  2. Symptoms include rash, itching, facial swelling, or breathing difficulty
  3. Stop the medication and seek immediate medical attention if allergic signs appear

Overdose Information:
  1. Overdose symptoms may include severe suppression of blood counts, vomiting, and arrhythmias
  2. There is no specific antidote for dasatinib overdose
  3. Supportive care and hospitalization may be required
  4. Inform your doctor immediately or visit the nearest emergency center if overdose is suspected

Missed Dose Instructions:
  1. If you miss a dose, skip it and take the next scheduled dose
  2. Do not double up the dose to make up for the missed one
  3. Maintain regular timing each day to ensure stable blood levels

Additional Notes:
  1. Dyronib is a long-term treatment; compliance is crucial for success
  2. Avoid activities that increase bleeding risk if platelet count is low
  3. Report symptoms like swelling, breathing issues, or unusual bruising promptly
  4. Periodic monitoring for heart and lung complications is necessary
  5. Keep regular follow-ups with your oncologist to adjust therapy if needed
